چکیده

Beginning in 1967, our laboratory has published a number of investigations of the nonsyngeneic stem ceil-inactivation phenomenon (1-3). I t has been shown that, after transplantation of a mixture of allogeneic lymphoid and hemopoietic cells into lethally irradiated hosts, stem elements of the mixture are inactivated by allogenic lymphocytes of the graft. In other words, after transplantation of such a cell mixture exogenous colony-forming units (CFU) ~ give no colonies in recipient spleens. On the basis of this fact we supposed that lymphoid cells of parental genot)~pe, being transplanted into sublethally irradiated F1 recipients, must inactivate host endogenous CFU, i.e., inhibit endogenous colony formation. The degree of inhibition may serve as a quantitatively accurate value for graft-versus-host (GVH) reaction. I t is evident that such an experimental system is characterized by the development of two simultaneously occurring processes: multiplication of endogenous CFU and homograft reaction against them by transplanted lymphocytes. Both processes may be quanti tat ively estimated by counting the number of spleen colonies in animals of corresponding experimental groups. By this technique, it is possible to compare quantitatively two of the most important actions of immunosuppressive agents: their mitostatic effect (CFU inhibition), and their lymphotoxic action; the latter is a true immunosuppressive effect, the abolition of endogenous CFU inactivation.

The antitumor effect of lowered doses of cytostatics combined with new analogs of vitamin D in mice.

Active and less toxic vitamin D analogs could be useful for clinical applications. In the present study, the antitumor effects of two new synthetic analogs of vitamin D, namely PRI-2202 (24R calcipotriol) and PRI-2205 (5, 6-trans calcipotriol), were evaluated. Since the analogs PRI-2202 and PRI-2205 administered alone inhibited tumor growth only slightly, they were applied in a combined therapy...

Inhibitory effects of 5-fluorouracil and oxaliplatin on human colorectal cancer cell survival are synergistically enhanced by sulindac sulfide.

BACKGROUND COX inhibitors appear to be promising agents in combination with cytostatics in the treatment of colorectal carcinoma (CRC). The aim of this study was to compare growth inhibitory effects of cytostatics (5-fluorouracil, 5-FU; oxaliplatin) and COX inhibitor sulindac sulfide (an active metabolite of sulindac), given alone or in combination, on several CRC cell lines. MATERIALS AND ME...

Antiproliferative activity of vitamin D compounds in combination with cytostatics.

Calcitriol is a potent antiproliferative agent against various tumour cells in vitro. Here, the results of a study on vitamin D compounds (calcitriol's analogues PRI-1906 and PRI-2191) as potential agents in combined antitumour therapy in vitro are presented. Applying antiproliferative SRB and MTT assays, the growth inhibitory effects of the vitamin D compounds, applied alone or in combination ...

An Experimental System for the Simultaneous Estimation of Mitostatic and Lymphotoxic Effects of Immunosuppressants and Cytostatics

Sublethally (600 R) irradiated (CBA x C57BL)F(1) mice were grafted intravenously with parental lymph node cells in doses ranging from 0.2 x 10(6) to 12 x 10(6). The transplantation of these lymphoid cells leads to inactivation of the recipient's endogenous CFU (as measured by the diminution of the number of colonies registered on the 10th day after irradiation). A 50% inactivation was observed ...

Novel Anticancer Platinum(IV) Complexes with Adamantylamine: Their Efficiency and Innovative Chemotherapy Strategies Modifying Lipid Metabolism

The impressive impact of cisplatin on cancer on one side and severe side effects, as well as the development of drug resistance during treatment on the other side, were the factors motivating scientists to design and synthesize new more potent analogues lacking disadvantages of cisplatin. Platinum(IV) complexes represent one of the perspective groups of platinum-based drugs. In this review, we ...
